Ezetimibe - Interactions and Warnings


Ezetimibe selectively inhibits the uptake of cholesterol. No clinically significant drug-drug interactions have been noted with the use of ezetimibe; however, further studies are warranted. The long-term safety and the effect on cardiovascular morbidity and mortality of ezetimibe are unknown.

Ezetimibe inhibits cholesterol absorption and promotes a compensatory increase of cholesterol synthesis, followed by clinically relevant reductions in LDL and total cholesterol concentrations. Ezetimibe also reduces plasma concentrations of the noncholesterol sterols sitosterol and campesterol, suggesting an effect on the absorption of these compounds as well.

The coadministration of ezetimibe at doses up to 10 mg with simvastatin 10 or 20 mg daily caused a significant additive reduction in low-density lipoprotein cholesterol compared with simvastatin alone. Additional clinical studies to assess the efficacy and safety of coadministration of ezetimibe and simvastatin are warranted.
